高效液相色谱-蒸发光散射法测定苹果中可溶性糖的含量 被引量:45

Determination of the Soluble Sugars in Apple by High Performance Liquid Chromatography with Evaporative Light Scattering Detector(HPLC-ELSD)

摘要 为了快速定量检测苹果中可溶性糖的含量,建立了应用高效液相色谱一蒸发光散射检测器(HPLC- ELSD)测定糖的方法,确定了高效液相色谱分离条件。色谱条件为:色谱柱(APS-2HYPERSIL,4.6 mmID×250 mm,5μm);流动相为V(乙腈):V(水)=80:20;柱温为室温;流速0.8 mL/min;进样量5μL;ELSD的漂移管温度40℃,氮气流速1.5 mL/min。结果表明,在此条件下,果糖、葡萄糖、蔗糖的浓度分别与峰面积成很好的线性关系(r>0.999),检测限分别为80、90、80 ng)加标回收率在96.75%~103.65%,相对标准偏差在1.41%~1.89%。研究证明,该方法具有快速、简便、准确的特点,是测定果蔬中可溶性糖含量较为理想的分析方法。 A HPLC-ELSD method for quantification anhlysis of the soluble sugar in apple was developed under the chromatographic condition by using column (APS-2HYPERSIL, 4.6 mmID×250 mm, 5 μm) and a mobile phase was V (acetonitrile) : V (water) = 80:20, column temperature - room temperature,flow rate - 0.8mL/min, sample volume - 5μL. The temperature of drift tube was 40℃, flow rate of N2- 1.5mL/min. The results showed that the concentrations of fructose, glucose and sucrose revealed a strongly positive correlations with the peak area respectively and the detection limit ranged from 80ng to 90ng, The average recoveries were 96.75%-103.65% and the relative standard deviations were 1.41%-1.89%. The experimental results indicated that this mothod was rapid, simple, accurate and an ideal way to determine soluble sugar content in fruits and vegetables.
