摘要 目的探讨微波热疗对人乳腺癌阿霉素细胞MCF-7和阿霉素耐药细胞MCF-7/ADM的多药耐药蛋白表达和耐药性的影响。方法应用Real-TimePCR法检测微波热疗对P-糖蛋白(P-gp)、多药耐药相关蛋白(MRP)表达的影响和高效液相色谱法(HPLC)检测微波热疗对细胞内阿霉素浓度的影响。结果MCF-7/ADM细胞在微波热疗后4h和24h,P-gp、MRP表达量明显下降(P<0.01)。微波热疗以后肿瘤细胞内阿霉素(ADM)浓度有明显上升(P<0.01)。结论微波热疗抑制了耐药蛋白的表达,增加了细胞内的药物浓度,热疗可能是逆转化疗耐药从而提高化疗效果的一种有效手段。 Objective To study the effect of microwave hyperthermia on expression of multidrug resistance related (MDR) protein and intracellu/ar ADM concentration of MCF-7/ADM cells. Methods Breast cell line MCF-7 and its ADM- resistance cell line MCF-7/ ADM were treated with ADM and microwave hyperthermia. The levels of cellular multidrug resistance related protein (MRP),P-glycoprotein (P-gp) were detected by Real-Time PCR. The intracellular concentration of adriamycin was measured by high performance liquid chromatography. Results Protein levels of MRP and P-gp in MCF-7/ADM cells were higher than that in MCF-7 cells,which were markedly decreased in MCF-7/ADM cells after microwave hyperthermia. The concentration of adriamycin in MCF-7/ADM cells was significantly increased after microwave hyperthermia. Conclusion Microwave hyperthermia inhibited the expression of multidrug resistance related (MDR) proteins and increased the intraeellular ADM concentration, The results suggest that microwave hyperthenmia could be used as a method to overcome MDR.
