摘要
为了构建GTPVP32基因缺失的通用GTPV转移载体,试验根据山羊痘病毒疫苗株(GTPV-TY)的P32基因结构设计了2对引物,分别扩增得到与P32基因部分相邻的长度约为1kb的同源重组臂序列并插入到pGEM-Teasy载体上,通过SmaⅠ位点重新连接,并在此位点插入CMV启动子、多克隆位点、BGHpolyA信号以及LacZ基因。结果表明,试验成功构建了通用山羊痘病毒P32基因缺失转移载体pdP32,此转移载体为改造GTPV-TY株以及开发二价或多价基因工程疫苗提供了有力工具。
The sequences nearby the Goat pox vaccine strain( GTPV -TY) P32 genes were used as homologous recombinant arms about 1 kb were amplified by PCR with primer pairs, and then they were inserted into pGEM - T easy vector respectively. The universal Goat pox virus deleted P32 transfer vector pdP32 -LaeZ were constructed by relinked the homologous recombinant arms on the Sma I site, and CMV promoter, muhielone sit, BGH polyA, LacZ genes were inserted into the Sma I site. This recombinant transfer vector should be useful for the development of GTPV - TY gene engineering vaccine.
