摘要
目的:观察恐伤孕鼠对其仔鼠大脑海马区细胞凋亡的影响。方法:采用恐伤孕鼠的方法造模,受孕后的雌鼠用随机取数法分为正常组、模型组、补肾组,造模时间为整个妊娠期间。仔鼠出生后14天,取其大脑海马区组织用流式细胞术检测其细胞凋亡率;用免疫组化ABC法观察Bcl-2、Bax在大脑海马区的蛋白表达。结果:模型组仔鼠海马区脑组织细胞凋亡率较正常组和补肾组增加(P<0.05)。同时,模型组仔鼠海马区脑组织Bcl-2阳性表达数下降、Bax阳性表达数升高,与正常组和补肾组比较均有极显著性差异(P<0.01)。结论:恐伤孕鼠通过凋亡相关基因Bcl-2蛋白表达下降、Bax蛋白表达增加共同作用引起其仔鼠海马区脑细胞凋亡增加,从而影响到仔鼠脑的发育,而补肾治疗可逆转此效应。
Objective:This study was designed to investigate the effect on progeny hippocampus cell apoptosis caused by horror injuring pregnant rats Method : Using the method of horror injuring pregnant rats during gestational period, the pregnant rats were randomly divided into normal group, model group and bu Shen group. We observed the hippocampus cell apoptosis rate collected from each group neonatal rats 14 days after the postnatal day. while the expression of Bcl-2. Bax gene in the neonatal rats hippocampus was observed by the immunohistochemical method. Result: The cell apoptosis rate rised in model group compared with the normal and bu shen groups ( P 〈 0.05 ), The expression of Bcl-2 gene decreased, Bax gene rised in model group as compared to in normal and bu shen groups( P 〈 0.01 ) . Conclusion:Horror injuring pregnant rats may bring about abnormal brain development of progeny, of which the mechanism may be the fised cell apoptosis rate which caused by the expression of Bcl-2 gene decreased and Bax gene rised, while bu shen prescription can adjust it into normal.
