瘦素对小鼠体外胚胎发育率及胚胎移植后着床率的影响 被引量:1

摘要 观察瘦素对体外受精后胚胎发育和胚胎移植后着床率的影响。采用小鼠卵母细胞体外受精后胚胎培养及胚胎移植的方法研究仅受精时添加的瘦素对受精后体外胚胎发育的影响及胚胎移植后着床率的影响。结果表明:1)仅受精时添加不同浓度的瘦素对受精卵发育至四细胞胚、桑椹胚及囊胚的比率均无显著影响(P>0.05)。2)添加的瘦素剂量为10ng/mL及50ng/mL时胚胎着床率较高,当添加的瘦素剂量为100ng/mL^500ng/mL胚胎着床率未提高,两组差别有统计学意义(P<0.05)。一定水平的瘦素可以显著提高小鼠的体外胚胎移植后的着床率,高水平瘦素反而对着床不利。受精后至着床前各期胚胎的发育率与受精当时瘦素浓度无关。 Objective: To investigate the effects of leptin on embryo development and imbed rate after embryo transplatation in mice. Methods: Study the effects of leptin in various concentration on in vitro embryo development and imbed rate after embryo transplatation, using in vitro embryo culture and embryo transplantatiom in mice. Results: Addition of leptin to embryo culture media could not promot the development from 4-cell stage embryo to morula and blastocyst( P 〉 0. 05). Leptin at 10 - 50ng/mL concentrations caused a dose-dependent promotion, nevertheless Leptin at even higher concentrations (100 and 500ng/mL) only brought on the same promotion as what the Leptin at concentration of 50ng/mL did (P〉 0.05). Conclusion : Embroys development before imbed have no relation to leptin added only in fertilizantion, keptin in suitable concentration can make imbed rate rise significantly and leptin in higher concentration have adverse effects on in vitro embryo imbed.
