摘要
通过分离纯化卵泡抑制素受体,确定其受体类型,旨在为抑制素的作用机制研究奠定基础。采集30头长白猪垂体组织制备膜蛋白,用亲和层析法从该蛋白组分中分离、纯化出抑制素受体。SDS-PAGE结果发现该受体提取物中有6种蛋白,分子质量从14 ku到130 ku不等,表明抑制素受体可能不是特异性的单一受体,而是共受体。改良的ELISA结果证实,该受体提取物特异性活性为99 U/mg,能与抑制素结合,且与剂量呈正相关(R=0.993)。
The type of the receptor of follicular inhibin was identified through isolation and purifica- tion,which would be used as the base for further study on inhibin mechanism. The pituitary of 30 sows of Landrace breed was collected and the membrane protein was prepared. The receptor of inhibin was isolated and purified from the above membrane protein through affinity chromatography. The result by SDS-PAGE showed that the purified inhibin receptor were some proteins with marker from 14 ku to 130 ku,and no single receptor may exist, but co-receptors were seen. And then, the result also suggested that special activity of the receptor was 99 U/rag and that the receptor might bind to inhibin, further-more,it was related to dose (R = 0. 993)
