摘要 目的:研究蛇床子素诱导小鼠胚胎成纤维细胞(NIH3T3)凋亡及其机制。方法:体外培养NIH3T3,不同浓度的蛇床子素作用于NIH3T3,应用溴化四氮唑蓝(MTT)比色法和生长曲线法检测蛇床子素对NIH3T3增殖活性的影响。Hoechst 33342荧光染色后,荧光显微镜下观察细胞核的变化。琼脂糖电泳分析法观察NIH3T3的DNA降解断裂的情况。结果:蛇床子素能明显抑制NIH3T3的生长,浓度为2.5×10-4mol/L时,细胞生长受到显著抑制,2.5×10-4mol/L蛇床子素作用24 h即可明显诱导NIH3T3的凋亡。结论:蛇床子素可明显抑制NIH3T3的生长并可以诱导其凋亡。 OBJECTIVE To study the effect of osthole on NIH3T3. METHODS NIH3T3 were treated by osthole on different concentration in vitro. The growth inhibitory effects were observed by MTT assay and cell growth curve. The characteristic of apoptosis was detected under the fluorescent microscopes by the staining of Hoechst 33342, also by DNA gel electrophoresis. RESULTS When the concentration of osthole was 2.5 × 10^ -4 mol/L the growth of cell was inhibited remarkably. Osthole induced apoptosis of NIH3T3 at concentration of 2.5 × 10^-4 mol/L. NIH3T3 treated by osthole showed typical characteristics of apoptosis including chromatin condensation and DNA fragmentation. CONCLUSION Osthole can efficiently inhibit the growth of NIH3T3 and induce apoptosis.
