大黄素对血管平滑肌细胞周期调控蛋白CyclinD_1表达的影响

Effect of Emodin on Expression of CyclinD_1 of Human Vascular Smooth Muscle Cells

目的:观察大黄素对人血管平滑肌细胞周期时相和CyclinD1表达的影响。方法:取对数增长期的平滑肌细胞,采用MTT法观察大黄素对平滑肌细胞增殖抑制的有效浓度范围,求出IC50并干预细胞;然后分别用流式细胞仪和Westernblot法进行细胞周期时相和CyclinD1表达的测定。结果:与对照组比较,IC50时药物组G0/G1期细胞百分比升高,S期细胞百分比下降,CyclinD1表达高峰延迟,表达量下调,细胞周期受阻于G0/G1期。结论:大黄素在抑制平滑肌细胞增殖的过程中通过下调Cyclin D1表达,阻滞了细胞周期的进程;是一种有效的抗平滑肌细胞增殖的药物。

Objective：To observe the effect of emodin on CyclinD1 and cell cycle phase of human smooth muscle cells （HSMCs） cultured in vitro and study the mechanism of inhibition of emodin on the proliferation of HSMCs. Methods ：The inhibitory effect of emodin on VSMC proliferation was studied by using MTT colorimeter,the effective concentration range and IC50 were calculated,then the cell cycle of the two groups was analyzed by using flow cytometer and CyctinD1 expressions were examined by the techniques of Western blotting. Results：Compared to the control group, the percentage of the cells of drug group in G0/G1 phase increased , and that in S phase decreased; the protein expression summit of CyclinD1 was delayed, the dose of protein was decreased, and the cells transition from G0/G1 to S was blocked by emodin. Conclusion：The inhibition of expression of CyclinD1 may be one of the mechanisms of emodin which is a effective drug on the inhibition of VSMC proliferation.
