摘要
目的:观察脑出血大鼠心肌组织IL-1β蛋白表达的变化及扶正护脑胶囊的干预作用。方法:大鼠随机分为假手术组、模型组、扶正护脑组和安宫牛黄组,分别于术后6、24、72h 3个时相点取材,采用免疫组织化学法测定心肌组织IL-1β蛋白表达,通过图像分析测定IOD值,并与血清CK-MB、cTnI含量作相关性分析。结果:IL-1β阳性颗粒主要在心内膜下的心肌细胞和血管内皮细胞的胞浆中表达。模型组各时间点心肌组织IL-1β蛋白表达均显著升高,其IOD值亦明显升高,扶正护脑组和安宫牛黄组均能明显降低IL-1β蛋白的表达,降低其IOD值。IOD值与cTnI含量呈显著正相关。结论:炎性因子IL-1β参与了脑出血诱发心肌损伤的机制,扶正护脑胶囊通过抑制IL-1β的蛋白表达减轻脑出血后心肌的损伤。
Objective: To observe the effect of Fuzhenghunao Capsule on IL-1β Protein Expression of myocardial tissues in Rats with intracerebral hemorrhage. Methods : 72 rats were randomly divided into 4 groups, sham,model,fuzhenghunao and angongniuhuang.The protein expression of IL-1β in rat myocardial tissues was detected by the immunohistochemistiy method,and measure the integrated optical density(IOD) with image analysis. Explore the correction between the IOD of IL-1β and the content of CK- MB and cTnI. Results : The positive particles of IL-1β was in the cytosol of myocardial cell and vascular endotheliocyte near to the endocardium. The IOD in Model group were higher than that in Sham group. The IOD in Fuzhenghunao group and Angongniuhang group were lower than that in Model group. Correction analysis showed that the IOD of IL-1β in the myocardial tissues was related with cTnI content.Conclusion : Inflammatory response attend the myocardial damage. Fuzhenghunao capsule could protect the myocardial tissues by inhibiting the Protein Expression of IL-1β.
