摘要 以白桦树种子和腋芽为外植体,通过研究培养基中不同植物生长调节剂配比对外植体诱导、芽苗增殖和生根等环节的影响,筛选出适合白桦组织培养的培养基配方。结果表明:以MS为基本培养基,附加蔗糖30g/L、琼脂6g/L,适合于白桦种子萌发生长的植物生长调节剂配比为BA0.2mg/L+NAA0.05mg/L,而BA1.0mg/L+NAA0.05mg/L适合腋芽诱导,诱导率高达87.0%,而且苗形态好,长势强,BA1.0mg/L+NAA0.05mg/L的植物生长调节剂配比利于芽苗的增殖,繁殖系数可达到每4周4~5倍,适于芽苗生根的培养基为1/2MS+NAA0.05mg/L,平均生根数为4.5条,最多根数为8条,根系发达,移栽后成活率达90%以上。 This study took seeds and axillary buds of Betula platyphylla Suk as explants, the effects of different plant growth regulator on induction and multiplication and rooting of plantlet were researched. The optimal medium for Betula platyphylla Suk was selected. The main results were as follows: The basic medium was MS supplemented with sugar 30 g/L+agar 6 g/L,was the optimal medium. For seed induction was BA 0.2 mg/L+NAA 0. 05 mg/L, BA 1.0 mg/L +NAA 0.05 mg/L was the optimal medium. For axillary buds induction, and induction rate was 87.0%. The optimal muhiplication medium was BA 1.0 mg/L+NAA 0. 05 rag/L, the optimal medium for rooting was 1/2 MS+ NAA 0. 05 mg/L, and the average number of root was 4.5, then transplanted. The survival rate of them was above 90%.
