摘要
建立了高效液相色谱测定何首乌中磷脂类化合物的新方法。采用Nuclesil100—5色谱柱(250×4.6mmi.d.,5μm),以甲醇为流动相,流速0.8mL/min,等度洗脱。蒸发光散射检测器的气体流速1.8L/min,漂移管温度66℃,进样量15ML。四种磷脂类化合物的线性范围分别为:磷脂酰甘油(PG):0.0388~0.388mg/mL(r^2=0.9991),磷脂酰乙醇胺(PE):0.0500~0.250mg/mL(r^2=0.9996),磷脂酰胆碱(PC):0.0103~0.165mg/mL(r^2=0.9995),溶血磷脂酰胆碱(LPC):0.0103~0.343mg/mL(r^2=0.9995)。该方法应用于首乌中磷脂类化合物的分析,取得了较好的结果。
A high performance liquid chromatographic method was developed to determine the phospholipids in Polygonurn rnultiflorurn Thumb. Separation was performed on a Nuclesil 100-5 column (250 × 4. 6 mm i. d. , 5 μm) , using methanol as mobile phase with a flow rate of 0. 8 mL/min. The carrier gas flow rate and drift tube temperature of evaporative light scattering detection were set at 1.8 L/min and 66℃, separately. The injecting volume was 15 μL. The linear ranges for different phospholipids were 0. 0388-0. 388 mg/mL (r^2 =0. 9991) for phosphatidylglycerol(PG), 0. 0500-0. 250 mg/mL (r^2 =0. 9996) for phosphatidylethanolamine(PE), 0. 0103- 0. 165 mg/mL (r^2 = 0. 9995) for phosphatidylcholine (PC), 0. 0103 - 0. 343 mg/mL (re = 0. 9995) for lysophosphatidylcholine (LPC). The proposed method was successfully applied to the determination of phospholipids in Polygonurn multi florum Thumb samples.
