摘要 目的比较4种常用蛋白浓度测定方法的准确性及各自的影响因素。方法采用Lowry法、BCA法、磺基水杨酸沉淀法、Bradford法等4种常用蛋白浓度测定方法对几种样品进行对比试验。结果研究了蛋白质样品中的添加物对不同方法的影响,采用BCA法测定PEG-IL-6的蛋白浓度以及采用Bradford法测定变性液中的蛋白浓度不仅准确而且重复性好。结论确定PEG-IL-6的最佳蛋白浓度测定方法为BCA法,而包涵体变性液中的蛋白浓度只能用Bradford法测定。 Purpose To compare accurary and effects of four methods which are Lowry, BCA, Bradford and Sulfosalicylic precipitation. Methods Determining and analying the values of IL-6, PEG-IL-6, Endotoxin, BSA and denature sample by four methods respectively. Results All sorts of additives of protein samples have different effects on different methods. It was found that BCA' method to measure PEG-IL-6 and Bradford' method to inclusion bodies were not only accurate, but also more repeatable. Conclusion BCA'method was more accurate and more precise for determination of PEG-IL-6 content that others. Bradford' method was only used to determined the concentration of inclusion bodies.
