摘要 【目的】探讨高浓度葡萄糖、高浓度晚期糖基化终产物(AGEs)对人单核细胞系U937细胞活性氧(ROS)生成的影响。【方法】用高浓度葡萄糖联合高浓度AGEs刺激U937细胞,观察24h内不同时间点细胞ROS产量变化。【结果】无论在正常培养还是高糖环境,加入高浓度AGEs均能够显著促进U937细胞ROS产量,而同一高浓度的AGEs存在下,高浓度葡萄糖对U937细胞ROS产量影响并不显著【结论】高糖和高AGEs联合作用可以明显改变U937细胞氧化应激水平。高浓度AGEs是促进U937细胞ROS产量增加的有利因素。 [Objective]To study the effect of high glucose and high AGEs on the generation of reactive oxygen species (ROS) in human monocyte line U937 cells. [Methods]U937 cells were cultured with medium with or without high concentrations of glucose and AGEs. The level of ROS in cells was detected at different time points within 24h. [Results]Overproduction of ROS in U937 cells could be detected when high concentration of AGEs existed, no matter whether the medium contained high glucose. However there was no remarkably difference between the effects of high glucose and normal medium on the production of ROS in cells when high concentration of AGEs existed. [Conclusion] The co-effect of high glucose and high AGEs can obviously induce oxidative stress in U937 cells. High concentration of AGEs is a powerful factor on the increase of ROS level in U937cells.
