摘要
目的研究铁调素(hepcidin)对人成骨细胞(hFOB1.19)内钙离子和铁离子转运的瞬时影响以及钙离子与铁离子转运的相互关系。方法将培养的成骨细胞分别进行钙离子和铁离子荧光染色,在激光共聚焦扫描显微镜(CLSM)下选定细胞后,实验组加入铁调素,对照组加相同体积的双蒸水,连续观察细胞内的铁离子以及钙离子荧光强度的变化。结果实验组细胞内的钙离子荧光强度在加入铁调素后形成一个瞬时的钙波,钙离子荧光强度增幅最大约为34%;对照组钙离子荧光强度呈现一个缓慢下降的趋势,两者变化趋势有明显差异。实验组细胞内的铁离子荧光强度在加入铁调素后呈现缓慢下降的趋势,与对照组内的铁离子荧光强度变化趋势相类似,无明显差异。结论铁调素有促进Ca2+向hFOB1.19内瞬时转运的作用,对hFOB1.19内铁离子瞬时转运的作用没有明显的影响。铁调素促进Ca2+离子向hFOB1.19内瞬时转运是非铁离子依赖性的。
Objective To study the instantaneous influence of hepcidin on calcium and iron transportation of human osteoblasts cells (hFOB 1.19), and the relationship between calcium and iron transportation. Methods hFOB1.19 cells were stained respectively with calcium and iron fluorescein stain, the fluorescence intensity and was observed with confocal laser scanning microscope (CLSM). After D2 H2O in control group or hepcidin in experiment group was added, the fluorescence intensity changes of the calcium and iron were recorded. Results The fluorescence intensity change of calcium increased and formed a calcium wave (maximum 34%) in experiment group, which was decreased slowly in control group. The fluorescence intensity change of iron decreased slowly in experiment group, and there was not significant difference between the two groups. Conclusion The calcium instantaneous transportation was increased with the increase of hepcidin concentration outside hFOB 1.19 cells. The instantaneous influence of hepcidin on iron ion is not significant. The instantaneous influence of hepcidin on calcium transoortation was not iron-deoendent.
