摘要
目的观察维甲酸灌胃大鼠骨质疏松模型肝脏铁调素(hepcidin)基因表达和血清铁调素蛋白含量的变化。方法72只雄性SD大鼠分为模型组和对照组。模型组动物以维甲酸70mg.kg-1.d-1持续灌胃14d;对照组动物以等量蒸馏水灌胃14d。选定时间点获取肝脏组织和血清,RT-PCR法测定肝脏铁调素基因表达,ELISA法测定铁调素蛋白含量。结果模型组肝脏铁调素基因表达相对量在模型中期明显低于对照组,模型初期和模型后期相对量较对照组低;对照组肝脏的铁调素基因各阶段表达改变均无明显差异性。模型组血清铁调素蛋白含量初期略降低,中期降低有显著差异性,后期含量降低,但较中期升高,整体变化趋势类似肝脏铁调素基因表达变化趋势;对照组血清铁调素蛋白含量改变较少。结论铁调素在骨质疏松模型建立过程中的变化可能与维甲酸灌胃骨质疏松模型形成有相关性,研究铁调素可为进一步探讨骨质疏松提供新的思路。
Objective To observe the changes of liver hepcidin gene expression and concentration of serum hepcidin in SD rats model of osteopososis induced by intragastic administration of tretinoin. Methods Seventy-two male SD rats were divided randomly into experimental group(E) and control group(C). Rats in group E were treated with tretinoin intragastic administration for 14 days,and those in group C with distilled water instead of tretinoin as in group E. The expression of hepatic hepcidin gene was detected by RT-PCR and the concentration of serum hepcidin by ELISA at preset time points. Results The expression of hepatic hepcidin gene in group E at intermediate stage was lower than that in group C, but there was no significant difference at early or late stage, which was not significantly changed during experiment in group C. The serum hepcidin was decreased slightly at early and late stages and significantly at intermediate stage. The whole tendency of changes in serum hepcidin was similar to the changes of hepatic hepcidin gene expression, which was not significantly changed during experiment in group C. Conclusion The changes of hepcidin in the formation of osteoporosis model in SD rats might have a relation with osteoporosis. A study on hepcidin may provide a new way in understanding osteoporosis.
