摘要
目的:探讨粉防己碱(tetrandrine,TET)的体内外抗氧化作用及该作用与其心肌保护药理功能的关系。方法:测定TET的体外清除DPPH自由基能力以及对过氧化氢(H_2O_2)损伤的H9c2大鼠心肌细胞活力的影响。并利用皮下注射异丙肾上腺素(ISO),在大鼠体内造成心肌损伤模型,从死亡率和心电图两方面观察TET的心肌保护作用。再初步检测TET对大鼠左心室超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽(GSH)以及丙二醛(MDA)4个体内抗氧化能力指标的影响。结果:TET能够显著性地、呈剂量依赖性地清除体外DPPH自由基、降低H_2O_2对H9c2细胞的生长抑制率。并且,预先腹腔注射TET能够降低ISO引起的大鼠死亡率和心电图ST段电压上抬的幅度,初步检测后发现,TET亦能够升高大鼠左心室SOD、CAT和GSH水平,降低MDA含量。结论:TET具有良好的心肌保护功能,其作用可能与它抗氧化的能力有关。
Authors studied the in vitro and in vivo anti-oxidative effects of tetrandrine (TET) and the correlation between the eardioproteetive effects and anti-oxidative property. They evaluated free radical scavenging and anti-oxidative capacity of TET using DPPH assay and MTT assay, and its effects on H9C2 cells injured by hydrogen peroxide (H2O2). To investigate the eardioproteetive effects of TET, a rat model of myocardial injury induced by isoproterenol (ISO) was created, and the superoxide dismutase (SOD), eatalase (CAT), glutathione (GSH), and malonaldehyde (MDA) levels of left ventricle were roughly measured. It is apparent that TET can dose-dependently scavenge DPPH free radical, and block the growth inhibition induced by H2O2 on H9C2 cells. Tetrandrine based pretreatment brings down mortality, and suppresses ST segment elevation induced by ISO. TET also prevents the SOD, CAT, GSH down-regulation and the MDA up-regulation. Authors believe that TET has a prominent eardioproteetive effect that may result from its anti-oxidative capacity.
