复方丹参滴丸及其主要成分丹参、三七对缺血再灌注引起的大鼠肠系膜微循环障碍的多环节改善作用 被引量:7

摘要 目的:本研究探讨复方丹参滴丸(cardiotonic pill@,CP)及其主要成分丹参、三七对(is—chemia/reperfusion,I/R)引起的大鼠肠系膜微循环障碍的改善作用。方法:用连接于彩色摄影机、高敏感摄影机的倒置生物显微镜观察和纪录Wistar雄性大鼠肠系膜微循环的动态。将肠系膜动静脉同时结扎10min,然后解除结扎,建立I/R模型。假手术组(Sham)不做I/R处理。I/R处理60分前,Sham和I/R组灌胃给予生理盐水。CP+I/R组、丹参+I/R组、三七+I/R组分别灌胃给予CP、丹参水提物和三七水提物(各0.4g/Kg)。结果:丹参水提物可以抑制白细胞的粘附,抑制细静脉血管壁过氧化物,抑制FITC标记白蛋白的漏出。三七水提物可以抑制白细胞的粘附、肥大细胞脱颗粒。CP可以抑制白细胞的粘附、细静脉血管壁过氧化物、肥大细胞脱颗粒和FITC标记白蛋白的漏出。结论:CP多环节改善作用缺血再灌注引起的微循环障碍的作用是其所含有的丹参水提物和三七水提物相须作用的结果。 This is a study to understand the effects of cardiotonic pill@ (CP) and its major ingredients, Salviae Mihiorrhiza (SM) , and Panax Notoginseng (PN), on the microcirculatory disturbance induced by ischemia and reperfusion (I/R) in rat mesentery. In the study, researchers had a dynamic examination of microcirculation of mesentery of Wistar rats using an invert microscope equipped with a 3 charge-coupled device camera and a silicon-intensified target camera. I/R injuries were induced by ligation of the mesenteric artery and vein for 10 min, followed by reperfusion. Animals in the sham operated group received the same treatment, though without vessel ligation. Sixty min before I/R, saline was administrated by gavages for the animals in both I/R and sham groups, with CP (0.4g/Kg body weight), water-soluble extract of SM (0.4g/Kg body weight), and water-soluble extract of PN (0.4g/Kg body weight) for the animals in CP+I/R, SM+I/R and PN+I/R groups, respectively. CP was found able to ameliorate the I/R induced disorders in rat mesentery, including leukocyte adhesion, and the production of peroxides in venular walls, efflux of FITC labeled albumin across the venules, and mast cell degranulation. The water soluble extract of SM has a similar manifestation, except that it exerted no effects on mast cell degranulation. On the other hand, the water soluble extract of PN exhibited a significant attenuating effect on leukocyte adhesion and mast cell degranulation. It is believed that the multi-targeting potential of CP for attenuating the I/R induced microcirculatory disturbance in rat mesentery results from the coordination of the water soluble extract of SM and PN that it contains.
