摘要
背景与目的:探讨高电压脉冲电场诱导K562细胞凋亡的情况,寻求血液肿瘤治疗的新途径。材料与方法:采用高电压脉冲电场装置处理K562细胞,利用双标记流式细胞术(Annexin V-FITC/PI法)测定细胞凋亡率,通过透射电子显微镜观察K562细胞的超微结构。结果:经过高电压脉冲电场处理后的K562细胞,胞内出现大量空泡及裂隙,核内染色质向核周边聚集形成凋亡小体,呈典型细胞凋亡形态学改变;K562癌细胞的凋亡比例随着脉冲个数和电场强度的增加而增加。结论:高电压脉冲电场可以诱导K562细胞凋亡,该结果可为治疗癌症提供一个新的途径。
BACKGROUND AND AIM: A new approach to kill tumor cells in the blood through investigating apoptosis of K562 cells induced by high intensity pulsed electric fields. MATERIALS AND METHODS: Apoptosis of K562 cells was evaluated by transmission electron microscope and Annexin V-FITC and PI double labeled flow eytometry. RESULTS: There were many vacuoles and cracks in the cells, with heterochromatin margination, nuclear pyenosis and nuclear fragmentation. The high intensity pulsed electric fields caused damage in K562 cells, and the apoptotie percentage went up with the increase in frequency and intensity of PEF. CONCLUSION: K562 cell apoptosis was induced by high intensity pulsed electric fields,providing a new approach to kill tumors in the blood.
