摘要
目的:确定体外培养的成纤维细胞热休克对表达基质金属蛋白酶(MMP-1)和基质金属蛋白酶组织抑制因子(TIMP-1)的影响。方法:在体外培养的成纤维细胞热休克分别在37℃、43℃、45℃水浴30min,24h后应用ELISA法上清液检测MMP-1和TIMP-1表达,并与对照组比较。结果:体外培养的成纤维细胞热休克后,上清液中MMP-1明显增高,并呈温度依赖性。热休克对其表达TIMP-1无明显影响。结论:热休克早期成纤维细胞产生的MMP-1导致真皮基质降解,在皮肤光老化中有重要作用。
Objective: To observe the effect of heat shock on expression of matrix metalloproteinase - 1 (MMP- 1 ) and tissue inhibitor metalloproteinase- 1 (TIMP- 1 )in cultured human fibroblasts and investigate biological mechanisms of dermis after heat shock. Methods: Cultured human fibroblasts were treated by heat shock (37, 43, 45℃ ) and MMP- 1 and TIMP- 1 in the supematant liquid were measured by EHSA after 24 h. Results: The cultured human fibroblasts, which incurred heat shock in different temperature, increased MMP- 1 in the supematant liquid in a temperature- dependent manner. But heat shock did not affect the expression of TIMP- 1. Conclusion: MMP- 1 produced by early stage fibroblasts treated with heat shock, causing degradation of matrix in dermis, may play an important role in the process of photoaging of skin.
