摘要
以刺梨花药为试材,研究了不同预处理、附加物对刺梨花药愈伤组织形成及褐变的影响。结果表明:低温处理3 d,刺梨花药愈伤组织诱导率最高,褐化率最低;以100 mg/L维生素C浸泡处理2 h后,花药愈伤组织诱导率最高,为50.63%,而褐化率最低,为10.63%;在培养基中加入不同浓度的维生素C或硝酸银可以促进刺梨花药愈伤组织的形成,降低花药的褐化,其中在添加有100 mg/L的维生素C或50 mg/L的硝酸银的培养基中,花药褐化程度最低。
Using Rose roxburghii Tratt cv. anther as the materials, effects of factors, such as low temperature and vitamin C on callus induction from the anther and anther browning were studied. The results showed that after pretreatment at 4℃ for 3 days, callus induction rate was highest and browning rate was lowest. In the treatment with 100 mg/L Vc for 2 h , its induction rate was highest to 50.63% and browning rate was lowest to 10.63% . The medium supplemented with vitamin C or AgNO3 was advantageous to the induction of callus and reduced browning rate, especially the medium supplemented with 100 mg/L vitamin C or 50 mg/L AgNO3, its browning rate was lowest.
