高效液相色谱法测定糕点中富马酸二甲酯

Determination of dimethyl fumarate in pastry by high performance liquid chromatography

目的：建立糕点中富马酸二甲酯的高效液相色谱测定方法。方法：色谱柱为Ec lipse XDB-C185μm,4.6 mm×150 mm柱;流动相为甲醇-0.03 mol/L乙酸钠-0.03 mol/L冰醋酸-0.008 mol/L溴化四丁铵的水溶液混合液（40：60）。样品粉碎超声提取后,经0.45μm滤膜过滤,进样测定,保留时间定性,峰面积定量。结果：标准曲线线性良好,方法线性范围10～200μg/ml,相关系数r=0.9993,回收率为85.5%～93.5%,相对标准偏差为2.6%～4.6%。结论：该方法操作简便,具有灵敏、准确、无杂质干扰等优点,可用于糕点中富马酸二甲酯的测定。

Objective： To establish a rapid and sensitive high performance liquid chromatographic method for the determination of demethyl fumarate in food.Methods：Chromatographic column was Eclipse XDB-C18 5 μm,4.6 mm×150 mm.The mobile phase was methano 1：0.03 mol/L sodium acetate-0.03 mol/L acetic acid-0.08 mol/L BTBA（40：60）.Sample was crushed and used 0.45 μm membrane for filtration after ultrasonic extraction.And then injected into HPLC,the component was qualitatively determined by retention time and quantitatively determined by peak-area.Results：The standard curve showing a direct ratio between the concentration and the peak area were obtained（r=0.9993）.Method linear range was 10～200 μg/ml,the recovery percentages ranging from 85.5% to 93.5% and relative standard deviations were in the range 2.6%～4.6%.Conclusion：The method is proved to be satisfactory in precision,accuracy and sensitivity.It can be used to detect demethyl fumarate in food.
