摘要
在油料磷脂含量测定的样品预处理中,标准分析方法耗费的试剂较多,时间较长,操作复杂。微波消解技术可以较好地克服以上缺点,对测定磷脂的微波消解-分光光度方法进行了比较研究。确定了样品消解液组成体系,通过正交试验,确定的微波消解最优方案为:微波时间8min,微波功率900W,样品用量0.2g,HNO36.0mL,H2O22.5mL。通过与GB5537-1985中干灰化法比较,微波消解法具有较好的精密度和准确度。
In the determination of phospholipid in oilseed, the standard determination method has many disadvantages, such as more reagent consumption,longer time and complex operation. A method of determining phospholipid by microwave digestion-spectrophotometry was introduced. By orthogonal experiment, the optimal microwave digestion conditions were obtained as follows: sample 0. 2g, microwave power 900 W, time 8 min, HNO3 dosage 6.0 mL, H2 O2 2. 5 mL. Compared with dry-ashing method of GB 5537 -1985 ,the microwave digestion method has better precision and accuracy, and could overcome the shortcomings of the standard method.
