嗅成鞘细胞提取液对胆碱能神经元移植治疗大鼠阿尔茨海默病的影响 被引量:7

摘要 目的探讨嗅成鞘细胞(OECs)提取液对胆碱能神经元移植治疗阿尔茨海默病(AD)大鼠的影响。方法SD大鼠双侧海马注射Aβ1-40,建立AD大鼠模型。实验动物32只分为4组:正常对照组、空白对照组、人工脑脊液移植组、OECs提取液移植组,每组8只。运用行为学测试、NADPH-d组化、刚果红组织学染色结合积分吸光度测定等技术,观察、比较各组AD模型大鼠移植2个月后学习记忆能力、一氧化氮合酶(NOS)阳性神经元数以及老年斑的变化。结果①行为学测试:人工脑脊液移植组、OECs提取液移植组大鼠达到学会标准所需训练次数(TN),错误反应次数(EN),全天总反应时间(TRT)少和潜伏期(D),短于空白对照组(P<0.05);以OECs提取液移植组更为显著(P<0.05)。②NADPH-d组化反应:人工脑脊液移植组、OECs提取液移植组NOS阳性神经元数较空白对照组明显增多(P<0.05),且前两组之间差异有显著性(P<0.05)。③刚果红染色:移植组及空白对照组三组间差异均有显著性(P<0.05),以OECs提取液移植组SP-IA值最小。结论嗅成鞘细胞提取液能明显提高胆碱能神经元移植治疗阿尔茨海默病大鼠的效果。 Objective To study the effect of distilled fluid of olfactory ensheathing cells (DFOECs) on choliner- gic nerve cell transplantion for the treatment of Alzheimer' s Disease (AD). Methods Rat AD model was created by in- jection of Aβ1-40 into double hippecampus. Rats were randomly divided into the normal control groups, blank control ones, manpower cerebral spinal fluid (MCSF) transplanted ones and DFOECs transplanted ones. There were eight rats in each group. Two months after transplantion, change of memory recovery was examined by Y - labyrinth test. The total number of nitric oxide synthase positive neurons and senile plaques of hippocampus were examined by NADPH - d and Congo red histochemical staining. Result (1)Y - labyrinth test: TN, EN, D and TRT of MCSF transplanted group and DFOECs transplanted group significantly decreased compared to blank control group ( P 〈 0.05 ), especially the DFOECs ones ( P 〈 0.05) ; (2)NADPH - d histochemical staining : the total number of NOS positive neurons of MCSF transplanted group and DFOECs transplanted ones were significantly greater than that of the blank control group ( P 〈 0.05 ) ; There was significant differences between the two transplanted groups (P 〈 0.05 ). (3)Congo red histochemical staining: compared to the normal control ones, there were significant changes in SP - IA in other three groups ( P 〈 0.05), with DFOECs trans- planted group being the lowest. Conclusion The distilled fluid of olfactory ensheathing cells can greatly improve the effect of cholinergic nerve cell transplantation for treatment of Alzheimer' s Disease rat.
