摘要
目的探讨电针对肝纤维化大鼠肝组织Ⅰ、Ⅲ型胶原mRNA表达的影响。方法Wistar大鼠随机分为4组(电针治疗组、药物治疗组、模型对照组和正常对照组),应用四氯化碳诱导大鼠肝纤维化模型,电针治疗组采用电针肝俞、足三里治疗,药物治疗组采用秋水仙碱治疗。应用Masson染色观察肝组织胶原纤维增生情况,采用实时荧光定量PCR技术检测肝组织Ⅰ、Ⅲ型胶原mRNA表达的变化。结果与正常对照组比较,模型对照组大鼠肝组织胶原增生,Ⅰ、Ⅲ型胶原mRNA表达均明显增加(均P<0.01);与模型对照组比较,电针治疗组大鼠肝组织胶原面积明显减少,Ⅰ、Ⅲ型胶原mRNA表达均明显降低(均P<0.01)。结论电针肝俞、足三里能够下调肝纤维化大鼠Ⅰ、Ⅲ型胶原mRNA的表达,该作用可能是其抗肝纤维化的重要分子机制。
Objective To investigate the effects of electro-acupuncture (EA) on expressions of collagen type Ⅰ and type Ⅲ mRNA in rats of hepatic fibrosis. Methods Wistar rats were randomly divided into EA-treated group,medicine-treated group,model group and normal group. Rat models of hepatic fibrosis were made by carbon tetrachloride. EA-treated group was punctured at Zusanli (ST36) and Ganshu(BL18) with EA,medicine-treated group was treated with colchicine. The collagen fiber proliferation were observed under light microscope with Masson staining, the expressions of collagen type Ⅰ and type Ⅲ mRNA were detected by real-time fluorescence quantitative polymerase chain reaction(FQ-PCR). Results Compared with normal group, the collagen hyperplasy and expressions of collagen typeⅠ and type Ⅲ mRNA increased significantly in the model group(P 〈0.01 ). Compared with model group, collagen hyperplasy and the expressions of collagen type Ⅰ and type Ⅲ mRNA decreased significantly in the EA-treated group( P 〈0.01 ). Conclusion Puncturing Zusanli( ST36 ) and Ganshu( BL18 ) with EA could down-regulate the expressions of collagen type Ⅰ and type Ⅲ mRNA, which maybe the important molecular mechanism of EA in preventing and treating hepatic fibrosis.
