摘要
采用超声法提取白苏中的齐墩果酸和熊果酸,用反相高效液相色谱法分离和测定其含量。色谱柱为Kromasil C_(18)柱(4.6 mm×250 mm,5μm),甲醇-0.2%磷酸溶液(88:12,V/V)为流动相进行等梯度洗脱,流速0.8 mL/min,光电二极管阵列检测器,检测波长210 nm,柱温28℃。齐墩果酸在(0.916~8.244)μg,熊果酸在(0.832~7.488)μg范围内具有良好线性关系,相关系数分别为0.999 9和0.999 8,平均回收率分别为97.2%(RSD为2.07%)和96.9%(RSD为2.30%)。选定的色谱条件可精确检测白苏中齐墩果酸和熊果酸的含量。
Oleanolic acid and ursolic acid in Perillafrutescens (L.) Britton was extracted by ultrasonic extraction and determined by RP-HPLC with photodiode array detector, The method was applied with a Kromasil C18 column (4.6 mm×250mm,5μm) by equivalent elution using methanol-0.2 % phosphoric acid solution (88 : 12, V/V) as the mobil phase. The flow rate was 0.8 mL/min and the detective wavelength was 210 nm, the column temperature was 28℃. Under these conditions, the calibration curve of oleanolic acid was linear within the range from 0.916 μg to 8.244 μg with r=0.999 9 and the average recovery was 97.2% with RSD=2.07%; the calibration curve of ursolic acid was linear within the range from 0.832 μg to 7.488 μg with r=0.999 8 and the average recovery was 96.9% with RSD=2.30%. The method is feasible for determination of oleanolic acid and ursolic acid in Perilla frutescens (L.) Britton.
