山医群体近交系中国地鼠动情周期的观察及染色法比较 被引量:3

摘要 目的完善山医群体近交系中国地鼠动情周期的繁殖生理数据。方法通过阴道脱落细胞检查法,连续15d对15只雌性地鼠的阴道脱落细胞经直接观察法、HE染色法、美蓝和伊红染色法染色后进行比较观察选定较好的方法,然后测定其动情周期。结果经比较HE染色法较好,观察雌性山医群体近交系中国地鼠的动情周期平均为(91.2±23.02)h,可区分为4个时相,即动情前期(16.82±6.71)h、动情期(20.97±12.41)h、动情后期(25.52±17.12)h和间期(28.55±16.99)h。结论经比较HE染色法能较好地反应细胞及细胞核的形态,能准确地判定地鼠的动情周期,可为完善中国地鼠动情周期的繁殖生理数据提供可靠的理论依据。 Objective To consummate the data of estrus cycle in inbred line of Chinese hamster of Shanyi colony. Methods Methylene blue staining,Eosine staining and HE staining were used to observe the vaginal exfoliate ceils in 15 female hamsters for 15 d. Then estrus cycle was determined by the better method. Results By comparison, HE staining was a better method. The average estrus cycle of inbred line of Chinese hamster of Shanyi colony was(91.2 ± 23.02)h, and it was distinguished into 4 phases by HE: preoestrus(16.82 ± 6.71) h,oestrus(20.97 ± 12.41) h, metoestms(25.52 ± 17.12) h, intermediate stage(28.55 ± 16.99) h. Conclusion The results indicate that HE staining could be a better method to show the appearance of the vaginal exfoliate cell and its nucleus and exactly distinguish the four phases of estrus cycle of inbred line of Chinese hamster of Shanyi colony, which provide a dependable theory basis for consummating the data of estrus cycle in inbred line of Chinese hamster of Shanyi colony.
