摘要 目的研究caveolin-1(小窝蛋白1)在骨巨细胞瘤中的表达,探讨其在骨巨细胞瘤发生发展中的可能作用。方法应用免疫组织化学SP法检测53例骨巨细胞瘤和22例良性骨肿瘤中caveolin-1的表达,镜下观察caveolin-1在肿瘤组织中的分布,应用自动图像分析检测caveolin-1的表达情况。结果①caveolin-1表达于大多数骨巨细胞瘤的肿瘤细胞上,定位于细胞膜和细胞质,阳性细胞在肿瘤组织中呈散在分布,无一定规律。caveolin-1在良性骨肿瘤中表达微弱。②caveolin-1阳性表达率在骨巨细胞瘤和良性骨肿瘤中分别为54.7%和25.0%(P<0.05),caveolin-1在骨巨细胞瘤中高表达。③caveolin-1的表达在骨巨细胞瘤Ⅰ级和Ⅲ级差异有统计学意义(P<0.01),而在骨巨细胞瘤其他分级之间无统计学差异(P>0.05)。结论caveolin-1在骨巨细胞瘤发生发展中可能发挥重要作用,并可作为骨巨细胞瘤Jaffe分级的辅助指标之一。 Objective To study the expression of caveolin-1 in giant cell tumor of bone(GCT), and to investigate its role in the carcinogenesis and development of GCT. Methods Caveolin-1 expression was detected by SP immunohistoehemical method in 53 cases of GCT and 22 eases of benign bone tumor. The distribution of caveolin-1 in tumor tissues was observed under optical micro- scope,and the expression level of caveolin-1 was detected by auto-image analysis. Results (1)Caveolin-1 was mainly located in the cell membranes and cytoplasm of the most GCT's tumor cells, and positive cells of caveolin-1 in the tumor tissues were scattered irregularly. Caveolin-1 was expressed weakly in benign bone tumor. (2)The positive rate of caveolin-1 in GCT(54.7 % )was significantly higher than that in benign bone tumor(25.0 %, P 〈 0.05), and the expression level of caveolin- 1 in GCT was higher than that in benign bone tumor. (3)The expression level was significantly different between patients in grade Ⅰ and grade Ⅲ of GCT( P 〈 0.01 ), but not between patients in other grades(P 〉0.05). Conclusion Caveolin-1 may play an important role in the carcinogenesis and development of giant cell tumor of bone,and caveolin-1 may be one of auxiliary targets about Jaffe grade in GCT.
