摘要
目的通过对大鼠和家兔矽肺模型在高分辨率CT图像和病理结果的对照分析,确定家兔在复制矽肺模型上的可行性。方法通过气管内注射的方法将石英混悬液灌注入4只大鼠和4只家兔肺内建立矽肺模型。1只大鼠和1只家兔不处理作为对照。2个月后扫描动物即处死,取肺组织进行固定和切片HE染色。在光学显微镜下观察,将病理结果与影像表现作对照分析。结果灌注石英粉尘悬液后2个月,大鼠的HRCT图像仅能识别肺内大血管和叶、段级支气管结构,肺内的其他结构显示不清。家兔的HRCT图像显示,肺内点状密度增高影,边缘清晰,病理切片证实有早期矽结节形成。结论大鼠不适合作HRCT研究的模型动物;家兔肺模型提供了可靠的矽肺病变特征,因而可以应用到矽肺高分辨率CT的研究中。
Objective To determine the feasibility of rabbit as an experimental animal for establishing a silicosis model. Methods Four Wistar rats and 4 rabbits were infused with quartz dust suspension into tracheas to establish the silicosis model. One normal rat and one normal rabbit were chosen as the controls. All animals were executed at month 2 after infusion. Lung tissues were fixed and sliced after high resolution computerized tomography(HRCT)scanning. The imaging results were compared with the pathological results. Results At month 2 after injection of quartz dust suspension,HRCT images showed only main bronchi and larger vessels in rat model, with a poor quality. While shadow of heighten punctuate density was observed in rabbit model, with clear rim, and its earlystage silicotic nodule was confirmed by pathological .section. Conclusion Rabbit is suitable to be employed as an experimental subject for silicosis imaging study.
