祖师麻风湿膏对大鼠佐剂性关节炎的治疗作用

Therapeutic effect of Zushima Fengshi Gao on adjuvant arthritis in rats

目的观察祖师麻风湿膏对大鼠佐剂性关节炎的治疗作用,为其临床用于治疗类风湿性关节炎提供实验基础。方法采用容积法测定左、右后肢足跖容积后,将雄性Wistar大鼠50只,随机分为5组:正常组,佐剂性关节炎(AA)模型组,祖师麻风湿膏0.5g组,祖师麻风湿膏1.0g组和狗皮膏1.0g组(阳性药对照组),每组10只。正常组和AA模型组于后肢踝关节以下部位涂敷凡士林0.5g,其余各组贴敷相应药物。给大鼠右后肢足跖皮内注射完全弗氏佐剂每只0.1ml,制备AA大鼠模型(正常组每只注射生理盐水0.1ml),观察大鼠原发性病变和继发性病变情况,并对多发性关节炎症状进行评分。结果AA模型组大鼠原发性足肿胀度及继发性足肿胀度、多发性关节炎症状评分均较正常组明显增高,祖师麻风湿膏0.5g和1.0g组及狗皮膏1.0g组大鼠的原发性和继发性足肿胀度均较AA模型组显著降低(P<0.01),致炎后18h的肿胀抑制率分别为30.6%,36.9%和33.3%;非致炎侧继发性足肿胀的最大肿胀抑制率分别为38.6%,45.4%和40.9%,多发性关节炎症状评分也较AA模型组显著降低(P<0.05)。祖师麻风湿膏对大鼠佐剂性关节炎各指标的影响与阳性对照药狗皮膏组比较差异无统计学意义(P>0.05)。结论祖师麻风湿膏对实验性佐剂性关节炎具有一定的治疗作用。

Objective To investigate the therapeutic effect of Zushima Fengshi Gao（ZSMFSG）on adjuvant arthritis,and to provide an experimental basis for its clinical application. Methods The adjuvant arthritis（AA）rat model was established by injecting Freund＇s adjuvant 0.1 ml per rat into the right-hind paw. Fifty male Wistar rats were divided into 5 groups： normal group, AA model group, ZSMFSG 0.5 g and 1.0 g groups, and Goupi Gao（GPG） 1.0 g group（ positive control group）. ZSMFSG 0.5 g, 1.0 g and GPG 1.0 g were stuck to the hind limb of rats in the corresponding groups, respectively, while vaseline was smeared on those in normal group and AA model group. The primary and secondary swelling extents in hind paws were detected using volume testing method, and the .scores of polyarticular arthritis were also evaluated. Results Compared with normal group, primary and secondary hind paw swelling and the scores of polyarticular arthritis significantly increased in AA model group. Compared with AA model group, the indices decreased significantly in ZSMFSG 0.5 g, 1.0 g groups and GPG group（ P〈 0.01 ）. The swelling inhibition rates at 18 h after Freund＇ s adjuvant injection were 30.6 %, 36.9 % and 33.3 % in ZSMFSG 0.5 g, 1.0 g groups and GPG group, respectively, while the maximum inhibition rates of secondary swelling were 38.6 %, 45.4 % and 40.9 %, respectively. There was no significant difference in primary and secondary indices of adjuvant arthritis among ZSMFSG groups and GPG group. Conclusion ZSMFSG has therapeutic effect on experimental adjuvant arthritis in rats.
