摘要
目的利用超滤技术去除狂犬病疫苗生产过程中产生的细菌内毒素。方法采用截留相对分子质量30万的超滤膜对狂犬病毒收获液进行浓缩超滤,检测超滤后狂犬病毒浓缩液中内毒素残留量。结果用超滤技术去除狂犬病疫苗生产过程中产生的热原时,在适宜的压力下,狂犬病毒收获液浓缩80倍、洗脱25倍,细菌内毒素去除率可达87.5%~98.7%。结论采用截留相对分子质量为30万的超滤膜,以最佳运行参数进行去除内毒素的方法可以在工业化生产中推广。
Objective To eliminate endotoxin produced in the production course of rabies vaccine by ultrafiltration techniology. Methods The harvested liquid of rabies virus was concentrated and filtered by the ultrafiltration membrane of molecular mass cutoff 300 KD. After ultrafiltration, the residual amount of endotoxin in the concentrated solution of rabies virus was detected. Results In the course of eliminationg pyrogen in the rabies vaccine production by ultrafiltration, under proper pressure, the harvested liquid of rabies vaccine was concentrated by 80 times and eluted by 25 times. The eliminating rate of endotoxin was 87.5% -98. 7%. Conclusion To eliminate endotoxin by the ultrafiltration membrane of molecular mass cutoff 300 KD and optimum operational parameters can be popularized for expanded use in the industrial production.
