4个鸡品种TPK1基因的PCR-SSCP分析

摘要 以鸡硫胺焦磷酸激酶(TPK1)基因为候选基因,隐性白羽鸡、泰和乌骨鸡、萧山鸡、白耳鸡为试验材料,采用PCR-SSCP方法对TPK1所有外显子及3′-调控区进行单核苷酸多态性(SNP)筛查,探讨不同基因型与不同鸡品种肌肉硫胺素含量的关系。结果表明:在3′-调控区发现了1个SNP位点,其纯合子AA型与BB型相比有一个C2132T的单碱基突变,CC型与BB型相比有一个C2132A的单碱基突变。等位基因A和B的频率极显著高于等位基因C的频率,且泰和乌骨鸡与白耳鸡差异显著,故推测所检测到的基因座与肌肉硫胺素含量相关。 This study was designed to investigate the single nucleotide polymorphism (SNP) of TPK1 gene in various breeds, including Recessive White chickens, Taihe Silkies chickens, Xiaoshan chickens and Baier chickens. The primers for all the 7 exons and 3'-UTR in TPK1 gene were designed and the SNP was detected by PCR-SSCP and DNA sequencing. One SNP was detected in the 3'-UTR. The sequencing results indicated that BB genotypes had only one base difference with AA genotypes C2132T, while CC genotype had only one base difference with BB genotype C2132A. The analytic results showed frequencies of allele B or A and allele C were significantly different (P 〈 0. 01 ), while Taihe Silkies chickens and Baier chickens were different (P 〈 0.05), it could conclude that the SNP may have relationship with thiamine content.
