摘要
应用光镜、电镜、荧光染色、DNA琼脂凝胶电泳等技术,系统地观察了顺铂处理后的人结肠癌细胞系SW1116的形态变化和生化特征。结果发现,癌细胞呈现染色质边集凝聚、胞浆皱缩等改变,符合凋亡细胞的形态学特点;凋亡细胞的出现具有时间和剂量的依赖性;在药物作用96h左右,检出“阶梯样”的DNA片段,这与血液系肿瘤细胞药物作用数小时就能检测到类似结果的报道不同,说明DNA寡核小体片段的出现并非结肠癌细胞系SW1116凋亡过程的早期事件。上述模型的建立,可用于深入研究细胞凋亡的发生机制。
We systematically observed the human colonic cancer cell line SW1116 after treatment with cisplatin in vitro. Under light,electron, fluorescent microscope, these cells showed morphological changes,including condensation of nuclear chromatin and cell shrinkage, which were indicative of apoptosis in a time and dosedependent fashion. After the drug acted about 96 hours, 'ladder' DNA fragments were detected in agar gel electrophoresis. This was different from previous report which described that similar findings could be seen when the drug acted on neoplasms of the blood system for several hours. Our finding suggested that the appearance of DNA oligonucleosome fragments wasn't an early event in the apoptosis process of colonic cancer cell line SW1116. It was consistent with the increase of floating cells. The slower rate and the longer duration of this new model for the induction of apoptosis was more convenient and suitable for the study of apoptosis mechanism. It was also suitable for the in-depth study of the relationship between apoptosis and regulated genes.
