颌面部鳞状上皮肿瘤DNA倍体及p53蛋白定量分析

摘要 采用荧光染色、双抗体标记染色和流式细胞分析技术,对55例颌面部鳞癌、乳头状瘤及正常面部皮肤组织进行DNA指数、增殖指数、S期细胞比例及p53基因产物表达的定量分析。结果:正常皮肤和乳头状瘤均为二倍体,而鳞癌异倍体率为60%,增殖指数和S期细胞所占的比例在这3种组织中均有明显的差异;在正常组织无p53阳性表达,乳头状瘤p53的阳性表达率在30%左右,鳞癌的阳性表达率为62.5%;鳞癌p53的阳性表达率随病理组织学分级的增高而增高,但与肿瘤细胞生物学特性无相关性。根据结果可以认为p53基因产物的表达在良恶性肿瘤组织中有非常显著性差异,并与肿瘤的恶性度有一定关系,因此,可将p53蛋白的检测作为良恶性肿瘤鉴别的辅助指标。 ? We analyzed the DNA index, proliferation index, cell ratio at S phase, and p53 gene product expression quantitatively, using fluorescence staining, dualantibodies's staining, flow cytometry analysis technique. The analysis included 55 cases of normal tissue, benign of malignant maxillofacial tumor initiated in squamous epithelium. Results: Normal sin and papiloma wer both dualpoidy. Aneuploidy ratio in squamous cacer was 60%. There was significant difference of the proliferation index and cell ratio at S phase in three kinds of tissues. p53 gene product was negative expression in normal tissue. The ratio of positive expression was 30% in papilloma, and 62.5% in squamous cancer. The ratio of p53 positive expression increased with the increasing of pathological grade in squamous cancer, but it was not correlated with biological characterization of tumor cell. These results showed that the expression of p53 gene product was significantly different between benign and malignant tumor, and was correlated with the degree of malignancy.
