牛黄解毒片经口对大鼠的急性和14天毒性

ACUTE AND 14 DAY REPEATED DOSING TOXICITY OF NIUHUANG JIEDU PIAN IN RATS

研究牛黄解毒片经口对大鼠的急性和14天毒性。方法：按卫生部药政局《中药新药研究指南（药学，药理学，毒理学）》的规定进行。结果：牛黄解毒片用6．43g／kg给大鼠一次灌胃，无死亡。持续14天给药，6．43g／kg和3.21g／kg剂量组大鼠体重增长缓慢，病理检查发现肝损伤。结论：牛黄解毒片经口对大鼠的急性LD_(50)大于6．43g／kg（临床用量50倍）；经口给药14天，毒作用阈值为3．21g／kg（临床用量25倍），靶器官为肝。

Objective: To examine the acute and 14 day repeated oral dosing toxicity of Niuhuang Jiedu Pian in rats Methods: Acoording to ' Guideline on study for new Chinese traditional drug (pharmaceutics,phamacology and toxicology), 1994'. Results: Wistar rats wer dosed by single gavage with Niuhuang Jiedu Pian at 6. 43 g/kg (equel to 50 times of clinical dose) and no rat dead. In the 14 day repeated oral dosing toxicity test, rats of 6. 43 g/kg and 3. 21 g/kg groups had a low increase in body weight and pathological liver damage. Conclusion: The oral LD50 of Niuhuang Jiedu Pian for rats was more than 6. 43 g/kg. In 14 day repeated dosing test in rats, toxicity threshold of Niuhuang Jiedu Pian was 3. 21 g/kg (equel to 25 times of clinical dose) and the target organ was liver.
