摘要
目的寻找与肝癌相关的新基因。方法应用Northern杂交技术对12个表达序列标记(expressedsequencetag,EST)克隆进行筛选,找到在肝癌及癌旁肝组织内有表达差异的基因,然后与基因数据库进行比较。结果Northern杂交结果显示,EST克隆F9391在所有6例肝细胞肝癌标本中呈高表达,而在相应的癌旁肝及2例正常组织内低表达。DNA序列测定并检索基因数据库GeneDatabases(EMBL96.6),表明该基因属于一未知新基因。结论F9391基因为一在肝癌组织内高表达的新基因,它与细胞的增殖相关,可能在肿瘤的发生中起重要作用,并且有可能成为肝癌诊治的一个新标志。
Objective Searching for novel gene related to human hepatoma. Methods Twelve EST (expressed sequence tag) clones were examined with Northern blot analysis, looking for the gene(s) differentially expressed in hepatoma versus surrounding noncancerous hepatic tissue, and then compared with Gene Databases. Results Northern blot analysis showed that EST clone F9391 expressed at high level in all of the 6 hepatocellular carcinoma samples, but at low level in the surrounding noncancerous hepatic tissues and in 2 cases of normal liver tissues. Nucleotide sequencing indicated that this clone belonged to a novel gene that had not included in the Gene Databases (EMBL 96.6). Conclusion F9391 gene is a novel gene overexpressed in hepatocellular carcinoma. Being related to cell proliferation, it might play an important role in liver oncogenesis. Forthemore, it may be used as a new marker for diagnosis and new target for treatment of hepatocelluar carcinoma.
