摘要
[目的]探究UV-C辐射对基因组完整性及蛋白表达的影响。[方法]CTAB法提取拟南芥基因组DNA,琼脂糖凝胶电泳检测基因组DNA的完整性。用TCA/丙酮法提取叶片总蛋白,通过SDS-PAGE凝胶检测蛋白表达变化。[结果]拟南芥基因组DNA随UV-C辐射时间的延长断裂明显增强、并诱导产生1条分子量约为66 kDa的蛋白条带。[结论]UV-C辐射可以引起拟南芥基因组DNA的断裂,对蛋白也有一定程度的影响。
[Objective] The research aimed to discuss the effects of UV-C radiation on genome integrity and protein expression of Arabidopsis thaliana.[Method] The genome DNA was extracted with CTAB method from Arabidopsis thaliana.The integrity of DNA was tested by agarose gel electrophoresis.Total protein was isolated from Arabidopsis thaliana with TCA/acetone method,and then tested by SDS-PAGE.[Result] The results indicated that the genome DNA fragmentation increased as the exposure time prolongation and a protein approximately 66 kDa was induced.[Conclusion] UV-C radiation could cause genome DNA fragmentation,and also had effects on protein expression.
