摘要 目的:建立高效液相色谱(HPLC)法,分离和测定紫苏子中色氨酸的含量。方法:采用Nova-Pak C18柱(150mm×3.9mm,4/am);流动相为0.01mol·L^-1醋酸和甲醇的二元梯度洗脱系统;流速为0.6mL·min;荧光检测器:激发波长为220nm;发射波长为355nm。结果:在选定色谱条件下其色氨酸的线性范围良好,样品加样回收率为96.7%~98.3%,RSD为1.17%~1.97%。结论:该方法精密度好,结果可靠,适合紫苏子中色氨酸的分析。 OBJECTIVE To establish an optimal HPLC method to determine the content of tryptophan in Perilla frutescens seeds. METHODS The content of this compound in the Perilla frutescens seeds were determined by reversed-phase high liquid chromatography. Nova-Pak C18 column(150 mm× 3.9 mm, 4 μm)was used, a mobile phase program of gradient elution with 0. 01 mol·L^-1 HAc with methanol at a flow rate of 0. 6 mL· min^-1 was employed with fluorescence detector: ex: 220 nm, em: 355 nm. RESULTS The good linear relationships were obtained under the optimum conditions. The recoveries of the stand- ards added were 96. 7% - 98. 3% while the relative standard deviations were within the range of 1.17% to 1.97%. CONCLUSION This method is accurate and reliable for the quantitive analysis of tryptophan in Perilla frutescens seeds.
