摘要
对蜜环菌进行分离纯化后筛选出性状最好的母种3号菌,利用0.5%的羟胺溶液对3号菌株的菌索处理2 min,诱变产生了生长速度快的突变菌株,该菌株比母种提前2 d萌发且菌索更加粗壮、密集。同时,研究了蜜环菌菌索在不同培养基上的生长特性。结果表明:在25℃、pH值5.5~6.0和暗培养的条件下,蜜环菌菌索在5号培养基上的生长速度最快,分枝多,密度大。通过一系列化学试剂对蜜环菌的作用,发现0.5%的羟胺溶液作用2 min后能够选育出生长速度快的优质蜜环菌,且遗传稳定。
By isolation and purification for Armillaria mellea, the NO.3 strain with the best characteristic was screened. The NO.3 strain was treated with 0.5% NH4OH in 2 minutes, a mutation strain grew quickly was selected, it bourgeon 2 days faster than initial strain. The rhizomorph was stronger and more intensive than initial strain. In the mean time, the growth characteristics of rihzomorph of Armillada mellea were studied on different culture media, the results showed the optimum conditions were 25℃, pH 5.5 - 6.0, in dark. Under the conditions, the strain with multiple branches and higher density grew fast on NO.5 medium. The author found that hy the treatment with 0.5% NH4OH in 2 minutes, the mutation strain grew quickly and had genetic stability.
