摘要
目的探讨芪棱汤对大鼠局灶性脑缺血再灌注损伤后微管相关蛋白-2(MAP-2)表达的影响及其神经保护作用机制。方法将大鼠随机分为正常组、假手术组、模型组、补阳还五汤组(对照组)及芪棱汤组(治疗组)。复制大鼠局灶性脑缺血再灌注模型并给予相应药物。在再灌注不同时点采用免疫组织化学法测MAP-2表达。结果正常组、假手术组可见MAP-2广泛表达;模型组、对照组及治疗组梗死中心区和周围区在再灌注即刻均出现表达下降,中心区随着再灌注时间延长持续下降,而梗死周围区在再灌注24h降至最低后表达有所上升;治疗组可增加MAP-2表达、促进其修复,以再灌注24h及48h明显,与模型组有明显差异,对照组在48hMAP-2表达亦增加,而治疗组效果更显著。结论芪棱汤能减少MAP-2丢失和促进其修复,具有良好的神经保护作用。
Objective: To study the nueral effect of of Qileng Decoction on MAP - 2 after focal cerebral ischemia - reperfusion injury in rats. Methods: The rats were randomly divided into 5 groups including the normal group, the sham - operation group, the MCAO(Middle Cerebrak Artery Occlusion ) group, the Buyanghuanwu Decoction group (the control gorup)and the Qileng Decoction group (the treatment group). The MCAO/Reperfusion model was established with method of internal careotid artery occlusion by suture. MAP-2 were observed with immunohistochemistry method at different reperfusion time. Results: MAP-2 was extenisvely expressed in the normal group and the sham-operation group. MAP - 2 positive cells decreased since the right reperfusion time both in the infarct central and surrounding region and reached to its minimun at 24h, and then increased at 48h in the infarct surrounding region. The MAP - 2 positive cells in the control group were more than that in the MCAO group dramatically at 48h while that in the treatment group were much more than that in both the MCAO and control group dramatically at 24h and 48h. Conclusion: Qileng Decoction has good effects against cerebral isehemia -reperfusion injury which may be due to its function of inhibiting MAP -2.
