摘要
目的研究氯氰菊酯和溴氰菊酯对雄性小鼠外周血淋巴细胞 DNA 的损伤。方法将64只清洁级健康雄性昆明种小鼠随机分为阴性对照组(花生油)、氯氰菊酯低(20mg/kg)、中(40mg/kg)、高(80mg/kg)剂量组,溴氰菊酯低(5mg/kg)、中(10mg/kg)、高(20mg/kg)剂量组和阳性对照组(环磷酰胺40mg/kg)。阳性对照组采用腹腔注射,其他各组采用灌胃染毒,10ml/kg,每天1次,连续染毒7 d。分别于实验前后测量小鼠体重。染毒后,取外周血淋巴细胞进行彗星实验。结果染毒前各组小鼠体重间比较,差异无统计学意义(P>0.05)。染毒后氯氰菊酯与溴氰菊酯高剂量组体重低于阴性对照组,差异均有统计学意义(P<0.05)。与阴性对照组比较,氯氰菊酯和溴氰菊酯高剂量组的拖尾率及拖尾长度较高,差异均有统计学意义(P<0.05)。且拖尾率及拖尾长度随着氯氰菊酯和溴氰菊酯染毒剂量的增大而升高,经线性趋势检验,差异均有统计学意义(P<0.05)。结论氯氰菊酯和溴氰菊酯对雄性小鼠外周血淋巴细胞具有一定的 DNA 损伤作用。
Objective To observe the damage of cypermethrin and deltamethrin to the male mouse lymphocyte DNA using comet assay. Methods Sixty-four male KM mice were divided randomly into eight groups:negative control group (peanut oil), eypermethrin low dose group (20 mg/kg), medium dose group (40 mg/kg), high dose group (80 mg/kg), deltamethrin low dose group (5 mg/kg ), medium dose group (10 mg/kg), high dose group(20 mg/kg) and the positive control group (cyclophosphamide 40 mg/kg). The mice from positive control group were exposed by intraperitoneal injection, the others were exposed by gavage for 10 ml/kg. Peripheral blood of mice was extracted after exposed for seven days, a comet assay was made with peripheral blood lymphocytes. Results Before the contamination, no significant difference of mice body weight were observed among the groups (P〉0.05). After the contamination, the mice body weight of high dose group of cypermethrin and deltamethrin were lower than the negative control group (P〈0.05). Compared with the negative control group, significant difference of trailing rate and the trailing tail length were observed above of the high dose group of cypermethrin and deltamethrin and the positive group (P〈0.05), high dose group of cypermethrin and dehamethrin and the positive control group result in comet rates rising and comets cell migration increasing of peripheral blood lymphocytes of mice. Linear regression analysis showed the dose of cypermethrin and dehamethrin were positively related with the trailing rate and the trailing tail length (P〈0.05). Conclusion Cypermethrin and deltamethrin has a certain damage role to DNA of peripheral blood lymphocyte of male mice.
