柚皮素抑制高脂饮食肥胖大鼠体重的实验研究 被引量:4

摘要 目的观察柚皮素对高脂饮食肥胖大鼠的影响并初步探讨其作用机制。方法SD大鼠44只随机分为正常对照组、高脂饮食组、高脂饮食+柚皮素低、中、高剂量组[柚皮素剂量分别为12.5、25、50mg/(kg·d)]。观察体重、热量摄入、血脂、血清游离脂肪酸(FFA)、血糖(BG)、胰岛素(In)、血清谷丙转氨酶(GPT)、谷草转氨酶(GOT)、丙二醛(MDA)含量和总抗氧化能力(T-AOC)的变化。结果给予高脂饮食后大鼠体重、热量摄入明显增加,BG和In浓度升高,FFA和低密度脂蛋白(LDL)增加,高密度脂蛋白(HDL)减少,血清GPT和MDA升高,但T-AOC降低。高脂饮食大鼠给予柚皮素后,体重减轻,热量摄入减少,BG、In、FFA降低,GPT和MDA减少,T-AOC增高。结论柚皮素可明显抑制高脂饮食大鼠的体重增加,并对肝细胞有一定的保护作用。其作用可能是通过改善体内脂肪代谢和胰岛素抵抗、提高机体的抗氧化能力实现的。 [Objective] To study the effect of Nafingenin on rats fed with high fat diet and discuss its mechanisms. [Methods] Forty-four healthy male SD rats were randomly divided into five groups: control group, high fat diet group,high fat diet plus low, medium and high dose of Naringenin group [The dosages of Naringenin are 12.5, 25 and 50 mg/(kg·d) respectively]. Food intake was monitored daffy,and body weight was weighed weekly at set time. Eight weeks later, the rats were sacrificed to collect blood for the measurement of the levels of serum lipid, glutamic pyruvic transaminase (GPT), glutamic oxalacetic transaminase(GOT), free fatty acid (FFA), blood glucose (Glu), insulin (Ins), malonaldehyde(MDA), total antioxidant capacity (T-AOC). [Results] Compared with control group, the rats in high fat diet group displayed increased body weight, caloric intake, the levels of Glu and serum Ins, FFA, GPT,low-density lipoprotein (LDL), MDA, reduced serum levels of high-density lipoprotein (HDL) and T- AOC. Naringenin lowered the body weight, caloric intake, the levels of Glu and serum Ins, FFA, GPT, MDA and elevated the levels of serum T-AOC in rats fed with high fat diet. [Conclusion] Naringenin can inhibit significantly body weight gain of high fat diet obese rats and exert protective effects on hepatocyte. Its mechanism may be due to the improvement of fat metabolism and insulin resistance and increasement of antioxidation activity.
