鸦胆子油对大鼠增生过长子宫内膜碱性成纤维细胞生长因子和血小板反应素-1表达的影响

摘要 目的检测不同浓度鸦胆子油在祛除大鼠增生过长子宫内膜中对碱性成纤维细胞生长因子(bFGF)、血小板反应素-1(TSP-1)表达的影响,明确其间的量效关系,探讨鸦胆子油治疗功能性子宫出血的机制。方法建立大鼠增生过长子宫内膜模型,分为正常对照组(A组),模型对照组(B组)及用药小、中、大剂量组(C、D、E组),通过免疫组化法检测各组中bFGF和TSP-1的表达情况。结果①bFGF:B组与A组比较有显著性差异(P<0.05);C、D、E组分别与B组比较有显著性差异(P均<0.05);C、D、E组间两两比较,均有显著性差异(P均<0.05)。②TSP-1:B组与A组比较无显著性差异(P>0.05);C、D、E组的表达程度较B组有增强趋势,但与B组比较无显著性差异(P均>0.05)。结论鸦胆子油对大鼠增生过长子宫内膜有祛除作用,可使大鼠增生过长子宫内膜中bFGF的表达减弱,对TSP-1表达无影响。 Objective It is to detect the expression influence of different concentration brucea oil on basic fibroblast growth factor (bFGF) and thrombospondin - 1 (TSP - 1) in dispelling proliferation excessively long endometrium of rats, to identify the dose- effect relationship and to discuss the mechanism of brucea oil on dysfunctional uterine bleeding. Methods The rat models of proliferation excessively long endometrial were established and divided into normal control group (group A), model control group (group B) and small, intermediate and big dosage medication group (group C, group D and group E). The expression situations of bFGF and TSP- 1 in the groups were detected with immunohistochemisty method. Results (1)bFGF: Group B had significant difference with group A (P 〈 0.05). Group C, group D and group E respectively had significant difference with group B (P 〈 0.05). Group C, group D and group E compared with each pair all had significant differences (P 〈 0.05). (2)TSP-1: Group B had no significant difference with group A (P 〉0.05). The expression degree in group C, group D and group E had enhancing tendency than group B, but there was no significant difference (P 〉0.05). Conclusion Brucea oil has dispelling action on proliferation excessively long endometrium of rats. It can weaken the expression of bFGF in proliferation excessively long endometrium of rats, but has no influence on the expression of TSP - 1.
