甘蓝型油菜新型恢复系Nsa恢1及其后代的GISH分析 被引量:3

摘要 用基因组原位杂交(genomicin situhybridization,GISH)结合双色荧光原位杂交(double-colour fluo-rescencein situhybridization,dc-FISH)技术,对甘蓝型油菜新型恢复系Nsa恢1及其后代进行了研究,结果表明,Nsa恢1为甘蓝型油菜-新疆野生油菜二体异附加系,包含了甘蓝型油菜全基因组和新疆野生油菜1对染色体,这1对新疆野生油菜染色体为同源染色体.不育系分别与保持系、恢复系Nsa恢1、保持系杂交后获得了BC3代不育株及可育株,GISH-dcFISH分析结果表明,BC3代可育株为甘蓝型油菜-新疆野生油菜单体异附加系,而不育株中不含新疆野生油菜染色体.说明恢复基因位于Nsa恢1的新疆野生油菜染色体上,为下一步克隆这一新型恢复基因提供了依据. The novel restorer line Nsa restorer-1 and its progenies were analysed by genomic in situ hybridization (GISH) combined with double-colour fluorescence in situ hybridization (dc-FISH) technologies, the results showed that Nsa restorer-1 was Brassica napus-Xinjiang wild Sinapis arvensis disomic alien ad- dition line, consisting of the whole genome of Brassica napus and a homologous chromosome pair of Xinjiang wild Sinapis arvensis. The sterile and fertile plants of BC3 generation, derived from the hybridization between sterile line and restorer line and maintaining line, were detected by GISH-dcFISH. The observations showed that the fertile plants were all the Brassica napus-Xinjiang wild Sinapis arvensis monosomic alien addition lines, and the sterile plants had no Sinapis arvensis chromosome. These results showed that the restorer gene was sited on the Sinapis arvensis chromosome pair of Nsa restorer-1, which will be a guide for our future research on the restorer gene cloning.
