摘要
从方法学上对氯化钙与甘油-聚乙二醇法制备JM105感受态细胞进行了比较。常规氯化钙法制备的细胞感受态仅维持3d(4℃条件下),而甘油-聚乙二醇法制备细胞其感受态可维持180d以上(-30℃条件下)。且两种方法制备的感受态细胞存放16h时其转化率分别为(6.42±0.06)×108和(1.73±0.03)×1010。
Two methods for preparation of competent JM105 cells were compared. The result showed that a higher transformation efficiency and a longer duration in competent condition were obtained with the 'glycerin-polyethylene glycol' method by comparison with the 'calcium chloride' method. The duration of competence was 180 days and 3 days, along with the transformation efficiency of (1.73±0.03)×1010 and (6.42±0. 06)×108 at 16 hours after being prepared, respectively.
