摘要
目的研究非诺贝特和肿瘤坏死因子α(TNF-α)对3T3-L1脂肪细胞内脂素(Visfatin)表达的影响。方法分化成熟的3T3-L1脂肪细胞,用不同终浓度非诺贝特(0、50、100、200μmol/L)刺激48 h;用100μmol/L非诺贝特刺激不同时间(0、12、24、48 h);用10 ng/ml TNF-α和10 ng/ml TNF-α加100μmol/L非诺贝特联合刺激48h。使用半定量RT-PCR法检测3T3-L1脂肪细胞Visfatin mRNA的表达。结果Visfatin mRNA表达随非诺贝特浓度增加、作用时间延长而增加;TNF-α作用48 h后Visfatin mRNA表达量减少(P<0.05),联用非诺贝特组高于TNF-α组(P<0.01)。结论非诺贝特能促进3T3-L1脂肪细胞Visfatin表达,这可能是其改善胰岛素抵抗的机制之一。
Objective To evaluate the effects of fenofibrate and TNF-α on visfatin mRNA expression in 3T3-L1 adipocytes. Methods The differentiated 3T3-L1 adipocytes were stimulated with fenofibrate at different concentrations for 48 hours or stimulated with 100 umol/L fenofibrate for different times ; The same cells were stimulated with 10 ng/ml TNF-α, mixture of 10 ng/ml TNF-α and 100 umol/L fenofibrate. The expression of visfatin mRNA was detected by RT-PCR. Resuits In 3T3-L1 adipocytes, with the increased concentrations of fenofibrate or the stimulated time extended, visfatin mRNA expression was also increased. Compared with control group, treatment with 10 ng/ml TNF-α had significantly decreased the expression of visfatin mRNA. Meanwhile, the expression of visfatin mRNA was increased significantly treatment with mixture of 10 ng/ml TNF-α and 100umol/L fenofibrate compared with 10 ng/ml TNF-α ( P 〈 0.01 ). Conclusion Fenofibrate increased the visfatin mRNA expression in differentiated 3T3-L1 adipecytes in a time-and-dose-dependent manner, which may one of the mechanisms underlying the action of fenofibrate on improving insulin resistance.
