摘要
目的:观察LPS诱导RAW264.7细胞内HMGB1转位及其释放。方法:用不同浓度的LPS处理小鼠RAW264.7细胞20 h,用荧光显微镜观察LPS诱导RAW264.7细胞HMGB1的分布情况。用Western blot方法检测HMGB1在胞核及培养上清中的水平。结果:LPS处理小鼠RAW264.7细胞20 h后,培养上清中的HMGB1水平明显增加,胞核HMGB1含量减少,并存在剂量依赖关系。结论:LPS能诱导RAW264.7细胞释放HMGB1,存在剂量依赖关系。
Objective:The purpose of this study was to investigate the translocation and release of HMGB1 in LPS - induced RAW264.7 cell. Methods:RAW264.7 cells were incubated with different concentrations LPS for 20 hours, the distribution and levels of HMGB 1 in RAW264.7 cells were subsequently analyzed by immunofluorence and Western blot, respectively. Results: HMGB1 accumulation in the culture media of LPS - induced RAW264.7 cells was significantly increased, and the accumulation of HMGB 1 in the nucleus was remarkably reduced. Conclusion: LPS can induce significant release of HMGB1 in RAW264.7 cells in a dose - dependent manner.
