摘要
目的:建立骨髓间充质干细胞与髓核细胞共培养的模型,研究间充质干细胞对体外培养的髓核细胞活性的影响。方法:通过transwell插入式培养皿构建间充质干细胞-髓核细胞共培养模型,观察共培养条件下髓核细胞形态变化,计数细胞增殖,ELISA方法检测蛋白多糖含量变化。结果:经过8 d共培养,实验组中的髓核细胞计数较对照组增加明显(P<0.01),培养上清液中蛋白多糖合成较对照组明显提高(P<0.01)。结论:成功建立共培养模型,与间充质干细胞共培养后髓核细胞活性上调。
Objective To establish a co-culture model of human nucleus pulposus (NP) cells and bone marrow-drived mesenchymal stem ceils (bMSCs) and to investigate its effect on the vitality of nucleus pulposus ceils. Methods NP ceils and bMSCs were co-cultured within transwell culture of 6-well plate. The NP ceils were counted for proliferation and proteoglycan was quantified in supernatants from the culture system using ELISA. Result The proliferation and proteoglycan synthesis of NP ceils were significantly greater after co-culture with bMSCs (P 〈 0.01 ). Conclusions The model for culturing NP ceils has been successfully established. The vitality of NP ceils is upregulated co-cultured with bMSCs.
