摘要
目的:建立裸大鼠人子宫内膜癌皮下瘤和宫腔原位移植瘤模型,初步观察其生物学特性。方法:体外培养子宫内膜癌细胞株Ishikawa,注射于8只雌性裸大鼠皮下,2周后处死2只成瘤裸大鼠,将皮下瘤修剪成2mm3的组织块移植入10只同种雌性裸大鼠左侧子宫内。4周后牺牲10只裸大鼠,取出左侧子宫。通过巨检、镜检、免疫组化、流式细胞仪等初步观察肿瘤的生物学特性。结果:8只雌性裸大鼠皮下均成瘤,10只雌性裸大鼠中7只左侧子宫内成瘤,免疫组化证实雌激素受体和孕激素受体在皮下瘤均有表达。流式细胞分析早期裸大鼠人子宫内膜癌的S期细胞含量为37.83%。结论:人子宫内膜癌裸大鼠皮下瘤和宫腔原位移植瘤模型成功建立,为子宫内膜癌的体内实验和新型治疗提供了较理想的动物模型。
Objective:To establish the subcutaneous transplantable tumor and orthotopic implanted tumor model of the human endometrial carcinoma in nude rat, and observe its biological features. Methods: Ishikawa cell line was cultured in-vitro and inoculated subcutaneously into eight female nude rats. Two weeks later, two nude rats were sacrificed and the subcutaneous tumors were excised. The subcutaneous tumors were cut into carcinoma mass with volume of 2mm^3 and then transplanted into the left uterine cavity of 10 nude rats. All ten nude rats were sacrificed 4 weeks after the transplantion. The left uterus were excised and the carcinoma mass were studied through histopathology, immunohistochemistry and flow cytometer. Results: The subcutaneous tumors of the human endometrial carcinoma were generated in all eight nude rats. The transplanted carcinoma was generated in the left uterine cavity in seven rats. The biological features of transplanted tumors were similar with the human endometrial carcinoma. The flow cytometer indicated the S-phase tumor cells accounted for 37.83 % in the human endometrial carcinoma of early stage in nude rats. Conclusion:The subcutaneous transplantable tumor model and orthotopic implanted tumor model using nude rat can serve as an ideal disease model in vivo study of human endometrial carcinoma and contribute to developing the new therapies of human endometrial carcinoma.
