维吾尔族宫颈癌基因组差异表达基因的层次聚类分析 被引量:2

摘要 目的:了解新疆维吾尔族宫颈癌患者基因表达谱的改变,探索新疆维吾尔族宫颈癌差异表达基因与其生物学特性的内在联系和规律。方法:收集我院2006年12月至2007年5月手术切除标本,分为A组(南疆维族妇女宫颈鳞状细胞癌标本7例)、B组(非宫颈病变宫颈手术标本4例),用21522个基因的Oligo芯片,经洗片、扫描、软件处理后,分析宫颈癌和正常宫颈标本之间基因表达谱的差异,同时通过数据处理,对本次实验与先前实验差异基因数据(C组)进行层次聚类分析。结果:宫颈癌组与正常宫颈组相比,差异表达基因1159条,以先前差异基因数据为基础,在检测数据中找到对应40条表达基因,显著差异表达基因17条,其中上调基因14条,下调基因3条。通过数据聚类分析,直观展示类内部个体在特征上有相似性。结论:通过两次实验集中找到了南疆维族妇女宫颈鳞癌极少部分差异表达基因,为进一步分析差异表达基因提供了实验依据。 Objective:To understand the changes of gene expression Uygur female with cervical cancer, to find the internal correlation between differentially expressed gene and bionomics. Methods :7 samples of cervical squamous cell cancer and 4 normal samples from Uygur female patients ,who lived in southern of Xinjiang Uygur Autonomy Region were collected during Dec. 2006 to May 2007. The human expression chip which contained 21522 gene was used. After the chip was laundered, scanned and processed using software, the difference was analyzed between cervical cancer expression profiling and normal sample expression profiling was nanlyzed. Different gene data between this experiment and previous experiment was also analyzed. Results:Compared with normal gene group and based on different gene data of prior-experiment, there were 40 expressive gene and 17 significantly differenrial expression-gene, up-regulation gene in 14 and down-regulation gene in 3. The result of cluster analysis showed that individuals in a cluster had same characteristic,individuals in different cluster had significantly different characteristic. Conclusion:Few differentially expressed genes were found in female Uygur patients, which can offer experiment evidence by analyzing the differential expression gene.
