雌二醇对实验性自身免疫性脑脊髓炎大鼠炎性细胞凋亡的研究 被引量:1

摘要 目的:探讨雌二醇(E2)对去卵巢后实验性自身免疫性脑脊髓炎(EAE)大鼠脑脊髓细胞凋亡的作用。方法:建立去卵巢10d后Wistar大鼠EAE模型,随机分为EAE组、E2治疗组和假手术组。E2治疗组从发病后连续3d,每日肌肉注射E2(1mg/kg)1次。在免疫后16d处死动物,取脑和脊髓用3′末端脱氧核糖核酸转移酶介导的duTP缺口末端标记(TUNEL)原位末端法检测大鼠脑脊髓凋亡细胞数。结果:与EAE对照组相比,E2治疗组和假手术组的中枢神经系统(CNS)TUNEL阳性细胞的凋亡率明显增高。结论:E2对EAE有治疗作用,其机制可能与其抑制CNS炎症细胞浸润及增加炎性细胞的凋亡有关。 Objective:To investigate the apoptosis of the inflamatory cells induced by estrogen in wistar rats with experimental allergic encephalomyelitis(EAE) which were ovariectomized.Methods:Wistar rats which were ovariectomized ten days were randomly divided into EAE group,E2 group and sham operated group.E2 group were injected with E2(1 mg/kg)once a day from postonset.On day 16p.i,the rats were killed to gain the brain and spinal cord.The apoptosis condition of spinal cords in rats was detected with in situ end-labeling method.Results:Compared with that of EAE group,apoptosis rate of TUNEL positive cells in E2 group and sham operated group in central nervous system(CNS) were obviously increased.Conclusion:E2 may exert therapeutical effect on EAE.This effect may be related to inhibit the infiltration of the inflammatory cells into CNS and induce apoptosis of this kind of inflammatory cells.
